Fully Human Monoclonal Antibody Preparation

A fully human antibody carries no foreign framework, which lowers the immunogenicity risk that comes with non-human sequences. To reach that outcome, the antibody is derived from a human repertoire from the start — either by screening a human antibody library or by generating the response in a host engineered to produce human immunoglobulin. Because the starting pool is already human, the selected lead needs little or no further humanization, and the work focuses on finding affinity and specificity rather than removing foreign sequence.

We prepare fully human monoclonal antibodies by building and screening human antibody libraries and, where suited to the target, by using a transgenic route that yields human immunoglobulin. The program moves from library to selected clone to confirmed binder, with the format chosen to match how the antibody will be used.

How a Fully Human Antibody Is Prepared

  • Establish the human repertoire – build a library from a human source or generate human immunoglobulin through a transgenic host.
  • Screen the library – pan the human scFv or Fab library against the target.
  • Recover and rank hits – sequence the enriched clones and assess binding.
  • Maturity the lead – improve affinity and developability through directed evolution where needed.
  • Reformat and confirm – express the candidate as scFv, Fab, or full IgG and verify performance.

Library and Source Options

Route What it provides
Human antibody library A diverse human scFv/Fab repertoire ready for phage display screening
Transgenic immunization Human immunoglobulin generated in a host engineered for the human response
Premade scFv / Fab pools Pre-existing human fragment libraries for accelerated screening

Performance Targets

  • Nanomolar affinity – selected leads reach the nanomolar range for binding and specificity.
  • Human sequence – the framework is human from the outset, minimizing later humanization.
  • Format flexibility – deliver as scFv, Fab, or full IgG per the application.
  • Documented selection – sequence and binding evidence accompany the candidate.

Applications

  • Therapeutic lead discovery – fully human candidates with low immunogenicity risk.
  • Diagnostic reagents – human binders for assay and detection formats.
  • Research tools – specific, human-sequence antibodies for study.
  • Further engineering – a clean human starting point for affinity maturation or reformatting.
